2、gNaSupervisor:ZhangNaAssociateProf.HuLiandongAcademicDegreeAppliedfor:MasterofScienceSpecialty:PharmaceuticalAnalysisUniversity:HebeiUniversityDateofOralExamination:June,2012摘要摘要本文以壳聚糖(CS)与羟丙甲纤维素(HPMC)为原料,采用简单的物理混合法制备了温敏性水凝胶。考察了溶液pH值、聚合物加入量、碳酸氢钠浓度等因素对凝胶形成温度的影响,确定了适宜的成胶条件。实验测定了CS/HPMC水凝胶的溶蚀动力学数据,
3、确定了该水凝胶的溶蚀动力学的基本规律和特征。红外光谱(FTIR)测定结果表明:HPMC是物理性的缠绕于CS网络结构中,分子间没有发生化学反应。通过扫描电镜(SEM)观察,确定了CS/HPMC水凝胶的形态和结构。电镜结果表明:所制备的凝胶都具有多孔性,载药凝胶比空白凝胶的结构略微致密。在空白水凝胶的基础上,分别以水溶性小分子药物5-氟尿嘧啶(5-Fu)和难溶性药物葛根黄豆苷元(DZ)为模型药物,考察凝胶释药性能。药物释放曲线结果表明,两种药物的释放行为与载药量、水凝胶组分有关。采用几种经典的数学模型对药物释放曲线进行拟合,再根据拟合的结果对释药机理做出分析。关键词温敏性水凝胶壳聚糖羟丙甲
