3、能否增加或抑制与清除结核杆菌的免疫功能,并对免疫功能改变的可能机制进行初步研究。方法:1.取雄性SD大鼠70只,体重180±30g,随机分为四组,分别为喂养前10只、喂养一个月、喂养三个月、喂养三个月停喂一个月,后三组均20只,分为实验组(10只)和对照组(10只)。2.喂养前10只自由饲养,不做任何干预,另外三组实验组用利福喷丁胶囊灌胃,对照组用生理盐水灌胃,余喂养条件相同。3.用流式细胞(flowcytometry,FCM)+++-+检测CD3、CD4、CD8及NK(CD3CD161)细胞计数,用碳粒廓
4、清法检测巨噬细胞的吞噬功能。结果:1.实验组与对照组各指标比较中,喂养一月及喂养三月停喂一月时各指标差异均无统计学意义,喂养3月实验++组与对照组各指标比较中,两组间的CD3、CD8、廓清指数、平均吞噬++++率四指标之间的差异有统计学意义,两组间的CD4、CD161、CD4/CD8三指标差异无统计学意义。2.对照组各时间点与空白对照组比较中,++两组间CD3、平均吞噬率、廓清指数之间的差异无统计学意义,CD4、++++CD8、CD161、CD4/CD8在喂养一月、喂养三月,喂养三月停喂一月及空白组之间差异
5、均有统计学意义;3.实验组各时间点与空白对照组比较:++++CD3、CD8、CD4/CD8这三个指标在喂养一月、喂养三月,喂养三月停喂++一月及空白组之间差异均有统计学意义,CD4、CD161、平均吞噬率、廓清指数这4个指标在各时间点间差异无统计学意义。结论:1.本实验中,实验组与对照组各指标比较中,喂养一月及喂养三月停喂一月时各指标差异均无统计学意义,表明喂养一月时利福喷丁胶囊的药效未达到影响免疫功能的强度,喂养三月停喂一月,可能由于停药后药物在体内的量减少或停药后原有药物随体内代谢清除,对各指标的差异+
6、+无显著性的改变;2.喂养3月实验组与对照组比较中,CD3、CD8、廓++清指数、平均吞噬率四指标差异有统计学意义,实验组CD3、CD8、廓清指数、平均吞噬率的均数±标准差均比对照组增高,且各指标差异有统计学意义,表明随着喂药时间的延长,药物对个别免疫相关指标+有一定影响。3.对照组各时间点与空白对照组比较中,除CD3、平均++++吞噬率、廓清指数外,指标CD4、CD4/CD8、CD161、在喂养一月、喂养三月、喂养三月停喂一月及空白组之间差异均有统计学意义,该差异可能为样本含量小导致的抽样误差或实验操作误
