1、重庆医科大学硕士学位论文鼻咽癌中PCDH17基因启动子甲基化状态的研究姓名:胡琴申请学位级别:硕士专业:耳鼻咽喉科学指导教师:胡国华201205重庆医科大学硕士学位论文鼻咽癌中PCDHl7基因启动子甲基化状态的研究摘要目的探讨鼻咽癌(Nasopharyngealcarcinoma,NPC)qbPCDH17基因启动子区CPG岛的甲基化状态,以及其对PCDHl7mI斟A转录、蛋白表达的影响。探讨PCDHl7基因在鼻咽癌中所起的作用。方法运用半定量反转录PeR(reversetranscriptionpolyme
2、raseChainPCRRT-PCR)和甲基化特异性PCR(Methylation-specificPCR,MSP)检测5株鼻咽癌细胞株,人喉鳞癌细胞株(Fadu),正常鼻咽上皮永生细胞系(NP69)qbPCDHl7mRNA的表达情况及PCDHl7的甲基化情况。用MSP检测鼻咽癌及正常组织中PCDHl7DNACPG岛的甲基化情况。应用去甲基化药物处理细胞株后检测PCDHl7mRNA表达的变化及PCDHl7基因甲基化的改变。构建PCDHl7质粒转染细胞得到稳定细胞株后,应用细胞克隆形成实验及划痕实验检测PCD
