1、中图分类号:R28Y1405475云南中医学弦2O08届攻读硕士学位研究生学位论文学洋虫抗胃黏膜损伤的实验研究研究方向主药蕴堡皇廑旦导师姓名萱丕副麴堡二oo八年五月洋虫抗胃黏膜损伤的实验研究研究生闰洪滨导师曹东副教授中文摘要目的:筛选洋虫抗胃黏膜损伤的活性部位,并通过幽门结扎型和乙醇损伤型胃溃疡模型深入观察其药效。方法:1.通过抗利血平致小鼠胃溃疡试验,比较洋虫的95%乙醇、石油醚、乙酸乙酯、正丁醇、水五个不同提取部位的抗消化性溃疡活性强度,确定其抗消化性溃疡的主要活性部位。2.经口给予洋虫乙酸乙酯提取物(ethylacetateextractofEartianusdefinestoides
2、Chevr.EAEMD),观察其对小鼠的急性毒性作用。3.通过结扎大鼠幽门,制造胃溃疡模型,观察EAEMD对大鼠胃酸分泌量、胃液PH值和胃蛋白酶活性的影响。4.通过经口70%乙醇造大鼠胃溃疡模型,观察EAEMD对大鼠胃黏膜损伤指数(UI)和血清及胃组织上清液中SOD、MDA、NO三个生化指标的影响。结果:EAEMD、正丁醇、水和95%乙醇提取部位能够对抗利血平致小鼠胃溃疡,降低胃黏膜损伤指数,其中EAEMD活性最强。EAEMD能够明显降低幽门结扎大鼠胃酸分泌量和胃蛋白酶活性;能够明显降低乙醇致大鼠胃溃疡的胃黏膜损伤指数,提高血清和胃组织上清液的SOD活性,增加NO含量,降低MDA含量。EAE
