1、第三军医大学硕士学位论文RoleofRANTESinthebasalcelloftheratepididymisAbstractTherateofhumanfertilityhasbeendecliningthroughouttheworldoverthepastthreedecades.Consequently,infertilityisnowamajorhealthproblemthataffectsatleastonesixthofcouplesworldwide.Whilealargepercentageofthesecouplesareafflictedbymalerep
2、roductiondysfunction,theetiologyofthefertilityproblemremainsunexplainedformostofthesemen.Theetiologyofmaleinfertilityoftenincludespermlowmotilityandthesperm-eggbindingforcereduction.Spermatozoaacquirethesecrucialfunctionsintheepididymis,wheretheyundergotheirfinalmaturationandarestored.Epididym
3、isiscomposedofalargenumberofseminiferoustubulesincomposition,morphologyandfunctionhaveobviousregionaldivision,theinitialsegment,proximalcaput,distalcaput,proximalcorpus,distalcorpusandcauda.Fourcelltypesarepresentintheepitheliumliningtheepididymallumen.Eachcelltypecontributestotheestablishment
4、andregulationofauniqueluminalenvironment.AlowluminalbicarbonateconcentrationandlowpHmaintainspermquiescentduringtheirmaturationandstorageinthisorgan,andepididymalepithelialcellsworkinaconcertedmanner,togetherwithspermatozoa,toestablishandmaintainthisacidicluminalenvironment.Clearcellsexpressth
5、eprotonpumpingATPase(V-ATPase)intheirapicalmembraneofepididymisepithelium,andactivelysecreteprotons.Luminalbicarbonate(HCO3-)inducesV-ATPaseaccumulationinapicalmicrovilliinclearcellsofepididymisepithelium,viasAC-dependentcAMPproduction.HCO3-issecretedfromprincipalcells.ATPisreleasedintothelume
6、nfromspermandprincipalcells.Inaddition,theV-ATPaseisregulatedbyluminalangiotensinIIviaactivationofbasalcells,whichcanextendnarrowbodyprojectionsthatcrossthetight-junctionbarrier.BasalcellsthensecreteNO,whichdiffusesouttostimulateprotonsecretioninclearcellsviaactivationofthecGMPpathway.Thus,ane
7、laboratecommunicationnetworkispresentbetweenprincipalcellsandclearcells,andbetweenbasalcellsandclearcellstocontrolluminalacidification.Monitoringanddecodingthese“intercellularconversations”willhelpdefinepathophysiologicalmechanismsunder
