1、硕士论文高纯度茄尼醇的制备与表征摘要茄尼醇作为一种重要的医药中间体,是合成医用辅酶Qlo和维生素K2的关键原料。因其碳链较长、合成步骤多而无法进行人工合成,目前主要由烟草提取得到。本文采用超临界C02静态、动态、静动结合和二次静态四种萃取方式对烟叶中茄尼醇进行粗提取,发现二次静态萃取提取效果较好。通过正交实验,考察了第二次静态萃取的温度、压力、时间和夹带剂用量等因素对茄尼醇提取综合指标的影响。优化工艺条件为:以95%乙醇为夹带剂,液固比为5.0mL/g,60℃、45MPa下萃取1h。此条件下茄尼醇选择性、提取率和转移率依次为31.59%,3.81%和97
2、.69%。实验还采用改进索氏提取法提取茄尼醇粗品。以无水乙醇为提取溶剂,液固比为30mL/g,70℃下回流4h,茄尼醇选择性、提取率和转移率依次为26.03%、2.84%和72.82%。实验表明,与改进索氏提取法相比,超临界C02二次静态萃取具有显著优势。以石油醚.乙酸乙酯(4:1,v/v)混合溶液为淋洗剂,由含量为31.59%的茄尼醇粗品经柱层析分离得到含量为85.01%的茄尼醇,再经重结晶得到98.62%的高纯度茄尼醇。产品结构经瓜、1HNMR、13CNMR和ESI.MS表征,与目标产物3,7,11,15,19,23,27,3l,35.九甲基.2,6
3、,10,14,18,22,26,30,34_三十六烷九烯.1.醇的特征相符。采用超临界C02二次静态萃取法对烟叶进行粗提取,茄尼醇提取率和转移率均高于现有方法,工艺条件缓和且对环境友好,可替代传统的热回流提取;采用柱层析.重结晶联合法制备含量>/98%的高纯度茄尼醇,总收率为1.58%,可为工业放大提供很好的指导。关键词:高纯度茄尼醇超临界C02二次静态萃取柱层析重结晶液相色谱.质谱联用硕士论文AbstractSolanesol,allimportantpharmaceuticalintermediate,isthekeystartingmaterial
