3、verseasreasearchdatashowedthatmildstraincrossprotection(MSCP)wouldbethemosteffectivemanagementagainststempittinginareaswherevirulentCTVisolatesandToxopteracitricida(Kirkaldy)werecommonlydistributed.DuetothegenotypecompositionofsevereandmildstrainsofCTV,associatedwiththecontroleffectofmildstrain

4、,quantitativedectectionofthedifferentCTVgenotypesofthemildstrainwouldhelpfurtherstudyonthemechanismofMSCP.Inthisstudy,apairofprimersT3-4F/RweredesignedwithintheORFlaofT3genotype,andtheSYBRGreenIreal-timeRT-PCRdetectionsystemwagestablishedwithoptimizedreactioncondition.Andthen.itssensitivityandr

5、eproducibilitywereestimated.ArapidspecificSYBRGreenIRT-PCRmethodfordetectingofT3genotypewasestablished,andusedtomonitorthedifferentgenotypecontentofthemildstrainsCTI1andCT31infield,togetherwithreportedreal-timefluorescencequantimtivemethods,detectingT30,T36,VTgenotypes.Inthemainwhile,theeffecto

6、fMSCPWasevaluedinthefield州tllmildstrainsCTI1andCT31pre-inoculated.Thefollowingresultshavebeenachieved:1.TheSYBRGreenIRT—qPCRdetectionsystemwasestablishedwithoptimizedreactioncondition.Thissystemhadstrongspecificityandhi曲sensitivitytoT3genotypeCTV,whichwas100timeshigherthanthatofconventionalRT-P
