1、沙棘叶中多糖的初步研究分类:毕业论文200706摘要本实验首次探讨了沙棘叶中多糖的提取分离,测定其多糖的百分含量。首先用蒸馏水清洗干叶,以洗掉尘土、浮渣,去掉粗梗,剪细。适量石油醚反复脱脂,滤渣自然挥干,再用常温水浸泡一次,80℃热水浸泡两次,以提取水溶性物质,然后合并所有水提液,采用旋转蒸发仪在适宜条件(t=50℃,抽真空0.09Mpa)下浓缩溶液至适宜体积,sevage法反复操作去蛋白,最后用95%的乙醇使溶液达到不同的浓度而沉淀出多糖,优选出沉淀沙棘多糖的最优醇浓度。0-5℃冰箱中放置48
2、小时后,过滤时用无水乙醇,丙酮洗数次,真空抽干。粗产品重新溶解于适量水中,然后通过大孔树脂进行纯化,蒸馏水洗脱,洗脱液在PH值为8时,用20%的H2O2,50℃以下保温2h脱色,以保证产品的颜色,最后用95%的乙醇使溶液醇浓度达到80%以析出多糖,重复上面的析糖操作,最后保存产品于干燥处。用D(+)—葡萄糖作标准,苯酚-硫酸法制备标准曲线,求出线性回归方程,从而得到粗多糖的含量,由此也可以算出沙棘叶中多糖的百分含量。然后进行TLC法初步定性确定多糖成分,再进行核磁共振定性分析,即可检验其准确成分
