2、vironmentalBiotechnologyNationalCoreResearchCenter,GyeongsangNationalUniversity,Jinju,660-701,KoreaAbstract:Wepreviouslyreportedthatriceblastfungusorjasmonicacidinducedtheexpressionofricepathogenesis-relatedclass10(JIOsPR10)proteins(Kimetal.2003,2004).However,nofurtherstudieshavebeencarriedouttoex
3、aminetheexpression,localization,andenzymaticactivityofthisproteinineitherdevelopmentaltissuesorintissuesunderabioticstressconditions.Inthisstudy,riceJIOsPR10wasexaminedbyWesternblotanalysis,immunolocalization,andbiochemicalassays.WesternblotsrevealedthattheJIOsPR10proteinwasexpressedindevelopmenta
4、ltissues,includinginflowerandroot.Theproteinwasalsoexpressedunderabioticstresses,suchasoccursduringsenescenceandwounding.Usingimmunohistochemicaltechniques,wedeterminedthatexpressionofJIOsPR10waslocalizedtothepaleaofflower,intheexodermis,andinnerpartoftheendodermisoftheroot.Insenescencingtissuesof
5、leafandcoleoptiles,itsexpressionwaslocalizedinvascularbundles.TheRNaseactivityusingJIOsPR10recombinantproteinwasdeterminedandabolishedaftertreatmentwithDTTinanativein-gelassay.Totestthis,wecreatedJIOsPR10mutantproteinscontainingserinesubstitutionsofaminoacidsC81S,C83S,orbothandexaminedtheirRNaseac
6、tivities.TheactivityoftheC83Smutantwasdecreasedintheagarosegelassaycomparedtothewildtype.Takentogether,wehypothesizethattheJIOsPR10proteinpossessesRNaseactivitythatissensitivetoDTT,suggestingtheimportanceofthedisulfidebondingbetweencysteineresiduesandthatitmightplayaroleinconstitutiveself-defensem
