1、山茱萸总苷的提取和大孔吸附树脂分离纯化工艺考察宋春敬1,王艳1﹡,于顺禄2,王慧明3(1.天津中医药大学中药学院天津市中药化学与分析重点实验室,天津300193;2.天津市天津医院,天津300211;3.天津中医药大学第一附属医院,天津300193)【摘 要】:目的:研究山茱萸中总苷的最佳提取工艺及大孔树脂纯化工艺。方法:以山茱萸干燥成熟果实为材料提取山茱萸总苷,莫诺苷含量为评价指标,选择乙醇浓度、料液比、提取时间、提取次数为考察因素,采用单因素和正交试验优选提取工艺,并进行验证试验。采用最佳工艺提取后,上HPD-300型大孔吸附树脂,水洗后分别用10%,30%,50%,70%的
2、乙醇洗脱,考察大孔树脂富集、纯化山茱萸总苷的吸附性能和洗脱参数。结果:50%乙醇是山茱萸总苷的最佳提取溶剂,提取次数对山茱萸总苷含量的影响最显著。大孔吸附树脂纯化过程中山茱萸总苷富集于30%乙醇洗脱液部分,洗脱流速为1.5BV·h-1,洗脱剂用量为3BV,洗脱率达94.9%。结论:山茱萸中山茱萸总苷的最佳提取工艺是50%乙醇提取,料液比1∶8,提取时间3h,提取次数3次。采用大孔吸附树脂可较好地纯化山茱萸总苷。【关键词】:山茱萸;总苷;正交实验;提取工艺;大孔吸附树脂Studyonextractionandpurificationofcornusofficinalissaposi
