2、eticengineeringtechnology,thetransgenicapproacheshavebeentriedtoovercometheseproblems.Duetothelowtransformationandregenerationrate,afewofcasesoftransgenicsoybeansweresuccessful.Toestablisheffectivetransformationandregenerationsystemsarestillhotinthesoybeantransformation.Boronplaysanimpor

3、tantroleinmaintainingthestructuralandfunctionalintegrityofcellwallandcellularmembraneinplant.Inthepresentexperiment,theinfluenceofboricconcentrationinthemediumontheinductionofsoybeancallusandmultipleshootsaswellasthetransformationeffeciencyofagrobacterium-mediatedreportgene(GUS)wereinvas

4、tigatedbyculturingtheexplantfromseedlingsofJilinsmallseedNo.7soybean.ThetransientexpressionratesofGUSgeneexpressionwerecalculatedbasedonthehistochemicalmethodandthestabletransformedshootswerefurtherdetectedbyPCRandDotblotting,inordertoexploretheefficiencyofsoybeantransformationinasuitabl

5、eboricacidconcentration.Resultsshowedthatlowboronconcentrationhadnosignificanteffectonthestructureofcallusinashorttime,whilehighboronculturemedium(50~300mg/L)resultedinstructuresofcalluscellsloosen.Whentheboricacidishigherthan300mg/L,theexplantsandcouldnotformnormalcallusduetotheserioust

6、oxicity.Moreover,boricacidconcentrationwith5.0mg/Lto10mg/L,aroundthenormalconcentrationofboricacid(6.2mg/L),hadthehightestmultipleshootsinductionrates.Bothhighboronandlowboronmediumwouldinhibittheinductionofmultipleshootsofcotyledonarynode,especiallyathighlevelboronmultipleshootsinductio
