1、-Abstract2,3-Butanediol(2,3-BD)isanimportantbiobasedbulkchemical.Usingstalkastherawmaterialtoproduce2,3-BDisverymeaningfulforpromotinghighvalueutilizationofstalk.Thisarticleusedwheatstalkandcorncobastherawmaterialsandremovedligninfromthestalksbypretreatment.Celluloseandhemicel

2、lulosewhichwereobtainedbypretreatmentofstalkweredegradedintofermentablesugarssuchashexoseandpentoseviaenzymatichydrolysis.Then,thefermentablesugarswereconvertedto2,3-BDbyklebsiellapneumoniaephysiologicalmetabolismandfinally2,3-BDwasseparatedfromfermentationbroth.Firstly,pretre

3、atmentandenzymatichydrolysisofstalkwerestudied.Theoptimumconditionsofpretreatmentare:thealkalineH2O2solutioncontaining2%NaOHand1.5%H2O2,theratioofalkalineH2O2toethanolof1:1,theratioofsolidtoliquidof1:20,80oC,for3h.Theremovalratesofligninfromwheatstalkandcorncobreach91.0%and92.

4、8%,respectively.Theoptimumconditionsofenzymatichydrolysisareasfollows:temperatureof50oC,pHof4.8,substrateconcentrationof80g/L,cellulaseloadingof28U/g(substrate),β-Glycosidaseenzymeloadingof15U/g(substrate)andreaction80h.Areducingsugaryieldof87.6%wasachieved.Theanalysesofgaschr

5、omatography-massspectrumonhydrolyzateofwheatstalkandcorncobshowedthattheirmaincompositionsareglucoseandxylose.Sencondly,thefermentationconditionsof2,3-BDwerestudied.Intheconvertionofglucoseinto2,3-BDbyKlebsiellapneumoniae,theoptimumconditionsaretemperatureof30oC,initialpHof7.0

6、,shakingspeedof150rpm,timeof96handinoculationamountof8%.Whentheinitialglucoseandpeptonecontentsinmediumare120g/Land16g/L,2,3-BDconcentrationreaches61.78g/L.Theoptimumfermentationmediumsof2,3-BDproductionfromstalkhydrolyzateareasfollows:theinitialglucoseandpeptonecontentsinmedi

7、umof120g/Land6g/L,adding0.05g/LofCaCO3,keepingpHof6.0-6.5duringtheprocessoffermentation.Thehighestconcentrationofprotuctcanreach50.97g/Lundertheaboveconditions.Inbatchfermentation,thepropersupplementalcarbonandnitrogenratiois21.Afterfermentation,themaximumoutputis91.88g/L,whic
