1、Abstract2,3-Butanediol(2,3-BD)isanimportantbiobasedbulkchemical.Usingstalkastherawmaterialtoproduce2,3-BDisverymeaningfulforpromotinghighvalueutilizationofstalk.Thisarticleusedwheatstalkandcorncobastherawmaterialsandremovedligninfromthestalksbypretreatment.Celluloseandhemicellulosewhichwe

2、reobtainedbypretreatmentofstalkweredegradedintofermentablesugarssuchashexoseandpentoseviaenzymatichydrolysis.Then,thefermentablesugarswereconvertedto2,3-BDbyklebsiellapneumoniaephysiologicalmetabolismandfinally2,3-BDwasseparatedfromfermentationbroth.Firstly,pretreatmentandenzymatichydroly

3、sisofstalkwerestudied.Theoptimumconditionsofpretreatmentare:thealkalineH2O2solutioncontaining2%NaOHand1.5%H2O2,otheratioofalkalineH2O2toethanolof1:1,theratioofsolidtoliquidof1:20,80C,for3h.Theremovalratesofligninfromwheatstalkandcorncobreach91.0%and92.8%,respectively.Theooptimumconditions

4、ofenzymatichydrolysisareasfollows:temperatureof50C,pHof4.8,substrateconcentrationof80g/L,cellulaseloadingof28U/g(substrate),β-Glycosidaseenzymeloadingof15U/g(substrate)andreaction80h.Areducingsugaryieldof87.6%wasachieved.Theanalysesofgaschromatography-massspectrumonhydrolyzateofwheatstalk

5、andcorncobshowedthattheirmaincompositionsareglucoseandxylose.Sencondly,thefermentationconditionsof2,3-BDwerestudied.Intheconvertionofglucoseinto2,3-BDbyKlebsiellapneumoniae,theoptimumconditionsaretemperatureof30oC,initialpHof7.0,shakingspeedof150rpm,timeof96handinoculationamountof8%.Whent

6、heinitialglucoseandpeptonecontentsinmediumare120g/Land16g/L,2,3-BDconcentrationreaches61.78g/L.Theoptimumfermentationmediumsof2,3-BDproductionfromstalkhydrolyzateareasfollows:theinitialglucoseandpeptonecontentsinmediumof120g/Land6g/L,adding0.05g/LofCaCO3,keepingpHof6.0-6.5duringtheprocess

7、offermentation.Thehighestconcentrationofprotuctcanreach50.97g/Lundertheaboveconditions.Inbatchfermentation,thepropersupplementalcarbonandnitrogenratiois21.Afterfermentation,themaximumoutputis91.88g/L,whichaccountsfor86%ofthetheoreticalyield.Theutilizationrateofreduc
