1、福建医科大学硕士学位论文缺氧对大鼠器官型海马脑片tau蛋白的影响姓名:于天莲申请学位级别:硕士专业:神经生物学指导教师:赵小贞20090601缺氧对大鼠器官型海马脑片tau蛋白的影响中文摘要目的1.建立4周龄大鼠海马脑片培养模型。2.观察缺氧对4周龄大鼠海马脑片tau蛋白的影响,以期建立拟痴呆组织模型。方法1.体外微孔膜插件培养海马脑片,应用倒置相差显微镜、LDH检测试剂盒、免疫荧光组织化学法及共聚焦激光扫描显微镜观察培养情况。2.标准条件培养7天后,入缺氧培养箱分别缺氧30min,lh,1.5h,
2、2h,然后恢复标准条件培养6天,应用倒置相差显微镜观察培养情况,用免疫荧光组织化学法及共聚焦激光扫描显微镜对tau蛋白表达水平进行观察和统计分析。结果1.培养海马脑片逐渐变薄,细胞条带亮度逐渐减弱;培养12d内,培养液中LDH含量稳定,海马脑片存活良好;在培养过程中,神经细胞逐渐减少,星形胶质细胞逐渐增多。2.p-Tau(Ser214)蛋白与总蛋白Tau.5的荧光强度比值显示缺氧lh组在培养第13天(即缺氧第6天)比对照组明显增大(尸<0.05),Tau(Ab.214)蛋白与总蛋白Tau.5的荧光强
