1、摘要在现代社会,肥胖问题已经成为全球性的一个公共健康疾病,引起了人们越来越多的关注。脂肪的生长涉及多基因表达、多信号途径及网络式调控,过程极其复杂。MicroRNA(miRNA)是一类存在于真核生物体内,大小为18~25nt的非编码(non-coding)内源性小分子物质,在多种生物学过程,包括细胞增殖、细胞分化、细胞凋亡、以及脂肪的生成代谢中发挥重要作用。选用小鼠、大鼠和鸡胚等实验动物来研究脂肪的生成机制,已经为解决肥胖问题提供了一些依据。但是选用矮小型鸡与正常型鸡来作为实验动物对脂肪的生成机制进行研究还未见报道。为了研究这两个品种鸡之间脂肪生成的差别,我们选用7
2、周龄的矮小型鸡和正常型鸡做为实验动物,运用miRNA微阵列技术分析腹脂中miRNA的表达谱,芯片结果显示:miR-10b,miR-148a,miR-199-3p,let-7k在矮小型鸡中表达上调,正常型鸡表达下调,其中miR-10b达到极显著差异(P﹤0.05);miR-181a-5p,miR-2188在矮小型中表达下调,正常型鸡表达上调,达到显著差异(P﹤0.01)。分析与脂肪生成相关的差异表达的miRNA的功能,能为人类和伴侣动物的肥胖症等能量代谢综合征寻找新的治疗靶标以及调控家畜的脂肪组织发育从而改善肉质提供借鉴和参考。关键词:鸡miRNA脂肪12TheAna
