2、cemethodofextractionrateasindex.ThecrudeyampolysaccharidewasdeproteinatedaccordingtotheTCAandhydrochloricacidmethod.Usingdeproteinizationrateandpolysaccharideretentionrateasindex,optimizeandfilterthebestmethodofdeproteinization.Andthecrudeyampolysaccharideafterdeproteinizationwasd
3、iscoloredbyH2O2methodandremovedsmallmoleculesbydialysismethod.CYPNandCYPAwereseparatedbyDEAE-52cellulosecolumnchromatography,andtheirpurityandmolecularweightweredeterminedbyhighperformancegelchromatography.PolysaccharidemonosaccharidecompositionandmolarratioofCYPNandCYPAweredetect
4、edbyHPLC.Inaddition,thestructureofCYPNandCYPAweredeterminedbyIRandNMR.Finally,theinfluencebeteweenyampolysaccharideandbacteria(Bifidobacterium,Lactobacillus,Escherichiacoli,Enterococcusfaecalis)weretestedbytheturbidimetrymethod.ResultsWeobtainedtheoptimumextractionofyampolysacchar
5、idebycoldwater:ratioofmaterialtoliquid10.30:1;extractiontimeof12.26h;extractionofthreetime.Asthisextractioncondition,theextractionrateofCYPNis2.89%andcontentofpolysaccharideis57.43%.Theoptimumextractionofyampolysaccharidebylye:ratioof-1materialtoliquid10.23:1;extractiontimeof7.59h
6、;concentrationoflyeof0.48mol·L.Asthisextractioncondition,theextractionrateofCYPAis24.23%andcontentofpolysaccharideis45.66%.Concentrationoftrichloroaceticacid3%istheoptimumdeproteinizationofTCAmethod,thedeproteinizationrateis95.03%,thepolysaccharideretentionrateis24.19%.ThepH2.0ist
