6、·············50个人简历··········································································································51中文摘要水飞蓟宾诱导细胞自噬改善高果糖所致HepG2细胞脂质沉积的作用及机制摘要近年来许多研究表明,细胞自噬与个体发育、氧化性损伤保护、肿瘤细胞的恶性增殖及神经退行性疾病有关。最近的研究表明自噬与脂代谢有关,肝脏是脂代谢的关键部位,脂代谢异常会引起肥胖相关疾病,如非酒精性脂肪肝病(NonalcoholicFattyLiverDisease,
7、NAFLD)、2型糖尿病、代谢综合征等。水飞蓟宾是一种从植物中提取的黄酮类化合物,具有保肝、降血脂、抗肝硬化、提高机体免疫力、抗氧化、保护心肌等作用,在临床试验研究中表明水飞蓟宾可以改善肝脏脂质沉积,改善胰岛素敏感性,但具体机制不明。本研究拟以HepG2细胞为研究对象,研究水飞蓟宾对高果糖诱导HepG2细胞脂质沉积的改变及对自噬水平的影响,探讨水飞蓟宾改善脂质沉积及自噬水平的作用机制,为临床用药提供实验基础及理论依据。目
