1、毕业论文红酵母高细胞密度酵母多糖提取工艺39毕业论文摘要本实验采用的是CICC编号31229的红酵母,通过对红酵母的实验,了解红酵母的特性及生长特点,进而改善红酵母的培养条件及培养基来扩大红酵母的生长浓度。研究从红酵母中提取酵母多糖的提取工艺,确定酵母多糖提取方法。采用摇瓶培养的方法,通过碳源、氮源及主要营养与环境条件等对红酵母产菌量的影响试验,以及葡萄糖、蛋白胨、酵母膏三因素三水平的正交试验,获得了红酵母的最佳生长条件:葡萄糖2%,蛋白胨1.0%,酵母膏0,5%,磷酸二氢钾0.15%,硫酸镁0.01%,氯化钠1.0%,初始pH5.0,接种量10%,500mL三角瓶中培养基装量为
2、80mL,发酵温度为28℃,摇床200r/min。在此优化的培养条件下,海洋红酵母的产菌量可达9.7亿个/mL,比优化前发酵培养基产菌量(5.6亿个/mL)提高了73.2%,比单因素试验结果(8.1亿个/mL)提高了19.8%。研究从酵母中提取胞壁多糖的提取工艺。提取工艺路线为:酵母溶解—冻融一超声波破碎一碱溶一中和一沉淀一洗涤—烘干。通过正交试验对酵母破壁和碱溶条件进行优化。初步寻求提取工艺条件,多糖得率为19.4%,用苯酚一硫酸法测定多糖的含量为51.9%。关键词:红酵母高细胞密度培养条件优化酵母多糖提取工艺39毕业论文AbstractTheexperimentwasthro
