1、全流量大鼠体外循环模型的建立作者:朱斌MihaiV.Podgoreanu,G.BurkhardMackensen,叶铁虎黄宇光【摘要】目的建立可存活全流量的大鼠体外循环(cardiopulmonarybypass,CPB)模型。方法8只雄性Sprague-Dawley大鼠异氟烷吸入诱导麻醉后,经口气管内插管,接呼吸机行机械通气。进行尾动脉、腹壁尾侧浅动静脉和右侧颈外静脉血管穿刺置管等外科准备。经颈外静脉置入的多孔套管为静脉引流通道,以经尾动脉置入套管为CPB动脉灌注管道,血液经过特制膜氧合器进行氧合建立CPB并进行90minCPB灌注。实验动物均进行血气和血流动力学等基本生理指
2、标监测,并在CPB前、CPB后60min以及CPB后1周测定细胞因子(IL-6和IL-10)以了解CPB所致的大鼠全身炎症情况。结果成功建立了不输血全流量大鼠CPB模型,所有实验动物全部存活至7天以上。受试动物IL-6和IL-10在CPB后显著升高,一周后回降到术前正常水平。结论本实验成功建立了全流量大鼠CPB模型,CPB后大鼠出现明显的全身炎症反应,这为研究CPB并发症病理生理机制以及相应的应对策略提供可靠经济的动物模型。【关键词】体外循环;大鼠;模型 Abstract:OBJECTIVEToestablisharecoveryratmodel12offull-flowca

4、dveins,andrightjugularveins.Themultiorificedcathetercannulatedinjugularveinwasusedforvenousdrainage;the20guagecathetercannulatedintailarterywasusedforthearterialinflowfromtheCPBcircuitandbloodwasoxygenatedwithacustomizedmini-oxygenator.Ninetyminutescardiopulmonarybypasswasperformedontherats.
5、Bloodgasanalysisandbasicphysiologicalmeasurementsweremonitored.Plasmacytokines(IL-6andIL-10)weremeasuredbeforeCPB,60minutesafterCPBand1weekafterCPB,inordertoobservetheCPB-inducedsystematicinflammation.RESULTSCPBwassuccessfullyestablishedandallsubjectswererecoveredfromsurgeryandCPBandsurvived
6、morethansevenobservationdays.TheplasmaconcentrationofIL-6andIL-10wassignificantlyincreasedafterCPBbutreturnedtothepreviousnormallevelin7dayspostoperatively.CONCLUSIONArecoveryratmodeloffull-flowcardiopulmonarybypasswassuccessfullyestablished.12SystematicinflammationwasexpectedlyinducedbyCPB,
