1、VEGF在大鼠缺血再灌注损伤肝组织中的表达及其意义【摘要】目的:探讨血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)在缺血再灌注损伤肝组织中的表达及意义。方法:利用雄性SD大鼠建立部分肝血流阻断动物模型,肝血流阻断90min后恢复血流灌注,再灌注后1,6h处死大鼠收集标本。分别采用全自动生化分析仪、HE染色,比色法、ELISA法测定血清转氨酶(ALT,AST),肝组织病理改变,肝组织髓过氧化物酶(MPO)的活性以及VEGF蛋白的含量。结果:与假手术组相比,缺血再灌注组(I/R组)缺血肝脏MPO活性,肝组织内VEGF蛋白的含量以及ALT,
2、AST在再灌注1h即升高,且缺血再灌注6h时较1h明显升高;缺血肝脏病理切片可见大量炎症细胞浸润,6h炎症反应更为剧烈,部分肝细胞呈点状坏死。结论:VEGF蛋白表达的上调可能参与了大鼠肝脏缺血再灌注过程中肝脏的损伤。【关键词】肝脏;缺血;再灌注损伤;血管内皮生长因子 [Abstract]Objective:Toinvestigatetheexpressionofvascularendothelialgrowthfactor(VEGF)intheliverischemia/reperfusioninjuryandsignificance.Methods:ThemaleSDrats

4、heactivityofhepaticmyeloperoxidase(MPO).ThechangesofVEGFproteininischemicliverlobesweredeterminedbyELISA.Thelivertissuespathologicalchangeswereevaluatedinhematoxylinandeosin(HE)stained.Results:After90minofpartialliverischemia,theAST,ALTlevelinI/Rgroupwassignificantlyincreasedascomparedwiththe
5、sham-operated(SH)group,andAST,ALTlevelatthe6hreperfusionwasmarklyhigherthanthatatthe1hreperfusion.TheexpressionofVEGFproteinandtheMPOactivityinhepatictissueswereremarkablyup-regulatedinI/RgroupthanintheSHgroupatthetimepointof1hand6hreperfusion.Theywerehigherat6hreperfusionthanthatat1h.ForHEst
