2、文献标识码:A文章编号:1004—7034(2014)06—0030—04关键词:肝硬化;问充质干细胞;造血干细胞;小鼠;骨髓摘要:为了验证联合应用两种干细胞治疗肝硬化疾病的有效性,试验采用贴壁培养法分离小鼠骨髓间充质干细胞,并在体外单克隆纯化,采用免疫磁珠法分离脐带血造血干细胞,对造血干细胞进行无血清培养及生物学性状检测,然后用间充质干细胞联合造血干细胞附加肝细胞生长因子治疗Babl/c裸鼠肝硬化。结果表明:成功纯化培养小鼠骨髓问充质干细胞,体外无血清培养获得脐带血造血干细胞:肝细胞生长因子40ng/
3、mL、间充质干细胞5×10。个、造血干细胞5x10。个组作用于Babl/c裸鼠肝硬化模型,Babl/c裸鼠肝功能显著改善,丙氨酸转氨酶由198U/L下降至86U/L,天冬氨酸转氨酶由398.7U/L下降至288.6U/L。说明小鼠骨髓间充质干细胞能够在体外实现单克隆纯化,小鼠脐带血造血干细胞可以在体外无血清培养并大量克隆,干细胞对肝硬化疾病具有明显的治疗效果。StudyOnthetreatmentoflivercirrhosisbvthecombination0fmOllsebonemarr0w—der
