2、1995;revisedandaccepted23June1995AbstractThegeneencodingthespike(S)proteinfromtwogeographicallydistinctstrains(AmericanandBritish)ofcaninecoronavirus(CCV)wasclonedandsequenced.Thenucleotidesequencerevealedopenreadingframesof1443or1453aminoacids,respec-tively.StructuralfeaturesincludeanN-terminal
3、hydrophobicsignalsequence,ahydrophiliccysteine-richclusterneartheC-terminus,twoheptadrepeatsand29or33potentialN-glycosylationsites.PairwisecomparisonsofSaminoacidsequencesfromtheseisolateswithotherCCVstrains(InsavclandK378)revealedthatheterogeneity,foundmostlyintheformofconservativesubstitutions
4、,isdistributedthroughoutthecaninesequences.However,5variableregionscouldbeidentified.Similaranalysiswithfeline,porcine,murine,chickenandhumancoronavirussequencesrevealedthatthecaninesequencesaremuchmorecloselyrelatedtothefelineSproteinsequencethantotheporcineSproteinsequenceseventhoughtheyareall
5、fromthesameantigenicgroup.Moreover,thesequencesimilaritybetweenCCVisolatesandthefelinecoronavirus,felineinfectiousperitonitisvirus(FIPV)wascomparable.ExpressionoftheCCVorthetransmissiblegastroenteritisvirus(TGEV)Sgeneusingthevacciniavirussystemproducedaproteinoftheexpectedsizewhichcouldinduceext
