1、葡聚糖磁性载体在大鼠腹部皮瓣中靶向性[摘要]目的:探索外磁场作用下葡聚糖和交联葡聚糖磁性载体在大鼠腹部皮瓣的靶向性,为磁性药物防治同种异体大鼠腹部皮瓣移植后免疫排斥作用做准备。方法:取雄性Sprague-Dawley(SD)大鼠35只,体重250~350g,随机分为以下7组:①交联葡聚糖磁性载体局部输注,2400GS磁场作用1h组(n二5,此组为标准对照组);②葡聚糖磁性载体局部输注.2400GS磁场作用1h组(n=5);③交联葡聚糖磁性载体全身输注,2400GS磁场作用1h组(n=5);④交联葡聚糖磁性载体局部输注,OGS磁场作用1h组(n二5);⑤交联葡聚
2、糖磁性载体局部输注,6000GS磁场作用1h组(n=5);⑥交联葡聚糖磁性载体局部输注,2400GS磁场作用3h组(n=5);⑦空白对照组(n=5)o取大鼠腹部皮瓣,用原子发射光谱法测定皮瓣内铁元素浓度,间接反映磁性载体在组织中的分布。结果:⑴两种磁性载体比较(第①组与第②组),在大鼠腹部皮瓣内,交联1聚糖磁性载体组铁元素浓度245.2±6.84pg/g高于葡聚糖磁粒组168.8±8.70pg/g,P<0.05o⑵两种输注途径比较(第①组与第③组),在大鼠腹部皮瓣内,局部输注组铁元素浓度245.2±6.84pg/g高于全身输注组215.2±8.79pg/g,P
3、<0.05o⑶不同外界磁场强度比较(第①组与第④、⑤组),在大鼠腹部皮瓣内,6000GS组铁元素浓度388.4±10.99pg/g高于2400GS组245.2±6.84pg/g,P<0.05,后者又高于OGS组127.8±5.54pg/g,P<0.05o⑷不同外界磁场作用时间比较(第①组与第⑥组),在大鼠腹部皮瓣内,3h组铁元素浓度414.4±13.07pg/g高于仆组245.2±6.84pg/g,P<0.05o结论:在外界磁场作用下,葡聚糖磁性载体可以在大鼠腹部皮瓣内靶向聚集,并且其浓度与磁性载体的性质和输注方式有关,与外界磁场强度及其作用时间呈正相关。[关
