1、DOI:10.16571/j.cnki.1008-8199.2013.10.029医学研究生学报2013年10月第26卷第10期JMedPostgra,Vol.26,No.10,October,2013·1049·论著(基础研究)LC-MS/MS测定大鼠血浆中没食子酸丙酯浓度王逢春,王敏,李秀敏,高守红[摘要]目的没食子酸丙酯是从中药赤芍中提取的有效成分没食子酸经酯化反应而成的化学药物。文中建立LC(液相)-MS(质谱)/MS方法研究大鼠口服没食子酸丙酯后的体内药代动力学。方法采用LC-MS/MS测定大鼠血浆中没食子酸丙酯的浓度,以没食子酸乙酯为内标,血浆样品用乙腈沉淀蛋白,
2、色谱柱为AgilentZorbaxSB-C18(3.5μm,100mm×2.1mm,I.D.),流动相为甲醇-乙腈-10mmol/L醋酸铵内含0.1%甲酸水溶液(10∶25∶65,V/V/V),流速0.25ml/min,柱温30℃。结果没食子酸丙酯的线性范围为5.0~500.0ng/ml(r=0.9984),最低定量检测浓度达5.0ng/ml(S/N>10),提取回收率>75%,日内和日间精密度RSD<15%。结论本法操作简单,选择性好,灵敏度高,费时少,适用于没食子酸丙酯在大鼠体内的药代动力学研究。[关键词]没食子酸丙酯;LC-MS/MS;血药浓度[中图分类号]R917[文
