1、大鼠心脏多巴胺1受体与心肌肥厚的关系作者:高嵩丹,王瑾,赵荣瑞,支建明【摘要】目的采用腹主动脉狭窄建立大鼠心肌肥厚模型,观察大鼠心肌肥厚时心肌组织多巴胺1受体(DA1-R)反应性的变化,借以阐明心脏DA1-R与心肌肥厚的关系。方法实验分5组:假手术对照组(C组)、腹主动脉狭窄组(A组)、腹主动脉狭窄+倍他乐克组(A+B组);腹主动脉狭窄+倍他乐克+SCH23390组(A+B+S组);假手术+倍他乐克+FODA组(B+F组)。各组均于4周后测定大鼠心功能和左心室/体重比值(LVW/BW);另外还测定了A组心肌肥厚大鼠的心肌组织在FODA
2、作用后cAMP生成量的变化。结果(1)A+B组各指标变化与A组无明显差异;(2)A+B+S组的LVW/BW与心肌细胞横截面积的明显减小,与A组或A+B组相比差异均有显著性(P<0.05);(3)B+F组LVW/BW明显加大,形成了一定程度的心肌肥厚,与对照组相比P<0.05。(4)A组大鼠心肌组织在FODA作用后,其cAMP的生成量明显高于对照组(P<0.05)。结论心脏DA1-R参与了病理情况下心肌肥厚的形成,由压力超负荷引起心肌肥厚时,心脏DA1-R的反应性增高。【关键词】心肌肥厚;多巴胺受体;fenoldopa
