2、ckground:Thecastorbean(RicinuscommunisL.),amonotypicspeciesinthespurgefamily(Euphorbiaceae,2n=20),isanimportantnon-edibleoilseedcropwidelycultivatedintropical,sub-tropicalandtemperatecountriesforitshigheconomicvalue.Becauseofthehighlevelofricinoleicacid(over85%)initsseedoi
3、l,thecastorbeanseedderivativesareoftenusedinaviationoil,lubricants,nylon,dyes,inks,soaps,adhesiveandbiodiesel.Duetolackofefficientmolecularmarkers,littleisknownaboutthepopulationgeneticdiversityandthegeneticrelationshipsamongcastorbeangermplasm.Efficientandrobustmolecularm
4、arkersareincreasinglyneededforbreedingandimprovingvarietiesincastorbean.TheadventofmoderngenomicshasproducedlargeamountsofpubliclyavailableDNAsequencedata.Inparticular,expressedsequencetags(ESTs)providevaluableresourcestodevelopgene-associatedSSRmarkers.Results:Intotal,18,
5、928publiclyavailablenon-redundantcastorbeanESTsequences,representingapproximately17.03Mb,wereevaluatedand7732SSRsitesin5,122ESTswereidentifiedbydatamining.CastorbeanexhibitedconsiderablyhighfrequencyofEST-SSRs.Wedevelopedandcharacterized118polymorphicEST-SSRmarkersfrom379p
6、rimerpairsflankingrepeatsbyscreening24castorbeansamplescollectedfromdifferentcountries.Atotalof350alleleswereidentifiedfrom118polymorphicSSRloci,rangingfrom2-6perlocus(A)withanaverageof2.97.TheEST-SSRmarkersdevelopeddisplayedmoderategenediversity(He)withanaverageof0.41.Gen
7、eticrelationshipsamong24germplasmswereinvestigatedusingthegenotypesof350alleles,showinggeographicpatternofgenotypesacrossgeneticdiversitycentersofcastorbean.Conclusion:CastorbeanESTsequencesexhibitedconsiderablyhighfrequencyofSSRsites,andwererichresourcesfordevelopingEST-S
8、SRmarkers.TheseEST-SSRmarkerswouldbeparticularlyusefulforbothgeneticmappingandpopulations
