5、mCLCnumber:S514.035.3Doc唧entcode:AArticle:2095—1191(2012)02—0155—05甜高梁组培苗生根培养的优化葛玉红,谢鸿锴,韩佳宇(广西大学农学院,南宁530005)摘要:【目的】优化能源作物甜高粱组培苗的生根培养体系,为遗传转化奠定基础。【方法】利用不同基本培养基(MS、1/2MS)对组培苗生根的影响进行筛选试验,得到较有利于生根的基本培养基;在最佳培养基中添加不同浓度的植物激素(IBA、NAA、ABT),综合根的质量、生根率、生根时间、生根苗的长势等筛选最佳生根培养基配方。【结果】筛选出最佳生根培养基组合:北甜三号(BT
6、3)为1/2Ms+2.Omg,LIBA,辽甜三号(LT3)为1/2MS+3.0mg/LIBA,且根粗、苗壮,两品种的生根率存在差异。此外,未加激素的Ms和1/2Ms培养基均无组培苗生根。【结论】在1/2Ms上添加2.om玑IBA或3.0m∥LIBA分别为北甜三号、辽甜三号组培苗生根的最佳途径。关键词:甜高粱;组培苗生根;优化;植物激素;基本培养基Sweetsorghumiswjdelyconsjderedasoneofthemostoutstandingenergycrop.Itsphotosyntheticefficiencyis2or3“meshigherthanthat
