3、spreadtoHongKong,Vietnam,CanadaandSingapore.TheSevereAcuteRespiratorySyndrome(SARS)viruswasidentifiedtobeanewvirusthatbelongstothecoronavirusfamily(Drostenetal.,2003;Ksiazeketal.,2003).Unliketheothermemberswhichcausemildrespiratorysymptoms(Holmes,1990),theSARScoronavirusresultinfatalitiesrangingfro
4、m10to15%oftheinfectedpatientsdependingontheagegroup.Thereplicationcyclewassimilartotheothercoronavirusesinthatthelatentperiodwasabout6hoursincellcultureshowevertheproductionofprogenyviruswasveryefficient(Sturman&Takemoto,1972;Ngetal.,2003).Microscopyinvestigationwasundertakentoseeifanyobservedhostc
5、hangesinducedbythevirusinfectioncouldhelpexplaintheseverityofthedisease.MethodsandMaterialsTheTEM,AFMandSEMwereusedtostudythemorphologicalchangesintheinfectedcells.Immuno-TEMwasalsousedtolocalizeauniqueSARSvirusprotein3a(U274)inrelationtotheinducedmorphologicalchanges.Attemptsweremadetolookforvirus
6、localizationandinducedmorphologicalchangesintheautopsylungtissuesusingtheTEM.ResultsandconclusionsIninvitrostudiestheSARSviruswasfoundtoreplicateatgreatspeedandproducedhighnumbersofprogenyvirusparticleswithin12to15hourspostinfection.Theearlychangesentailtheappearanceofthelargevacuolesenclosinglarge
7、myelin-likemembranewhorls.Thesemembranescouldhavearoleintheearlyeventofreplicationastheyareseencloselyassociatedwiththeuncoatedvirusnucleocapsids.Thesemembranestructuresremainedprominentduringthefirst10hour
