3、withdiethylaminoethyl-dextran(DEAE-D).ArecentreportthatRCVsgrowwellinL2mousefibroblastcellswasconfirmedandexpanded.RCVinfectionofL2cellswassubstantiallyenhancedbytreatmentofcellswithtrypsinbutnotbytreatmentwithDEAE-D.PrimaryisolationofSDAVfromexperimentallyinfectedratswasaccom-plishedusi
4、ngtrypsin-treatedL2cells.Oneof13additionalcelllinestested(raturinarybladderepithelium,RBL-02)supportedgrowthofRCVs,andgrowthwasslightlyenhancedbyDEAE-D,butnotbytrypsin.TheserefinementsofinvitrogrowthconditionsforRCVsshouldfacilitatefurtherstudiesoftheirbasicbiologyandimproveoptionsforpri
5、maryisolation.IntroductionRatcoronaviruses(RCVs),notablysialodacryoadenitisvirus(SDAV)andPar-ker'sratcoronavirus(PRCV),infectlaboratoryratsathighprevalence,causeclinicallyandeconomicallysignificantdiseaseandinterferewithresearchusingrats[7].DiagnosticandexperimentalstudiesofRCVshavebeenl
6、imitedbythelackofareliablecelllineinwhichtogrowtheviruses[7].RCVshavetraditionallybeengrowninprimaryratkidney(PRK)cellsorinfantmousebrainbutthesetechniquesrequiretheuseofanimalsandarelaborintensive.Althoughmanycoronaviruseshavefastidiousgrowthrequirements,replicatingonlyinorganculturesor
