2、analyzethepotentialroleofallelicvariabilityofcarotenoidbiosyntheticgenesintheinterspecifidiversityincarotenoidcompositionofCitrusjuices.Thesecondobjectivewastodeterminethenumberofcopiesforeachofthesegenes.Sevencarotenoidbiosyntheticgeneswereanalyzedusin呂restrictionfragmentlengthpolymorphism(RFLP
3、)andsimplesequeneerepeats(SSR)markers.RFLPanalyseswereperformedwiththegenomicDNAobtainedfrom25Citrusgenotypesusingseveralrestrictionenzymes.cDNAfragmentsofPsy,Pds,Zds,Lcyb,Lcy-e,Hy-b,andZepgeneslabeledwith[R-32P]dCTPwereusedasprobes.ForSSRanalyses,twoprimerpairsamplifyingtwoSSRsequencesidentifie
4、dfromexpressedsequeneetags(ESTs)ofLcy-bandHy-bgenesweredesigned.ThenumberofcopiesofthesevengenesrangedfromoneforLcy-btothreeforZds.ThegeneticdiversityrevealedbyRFLPandSSRprofileswasinagreementwiththegeneticdiversityobtainedfromneutralmolec(iLarmarkers.GeneticinterpretationofRFLPandSSRprofilesoff
