2、料重量的10倍浓缩,即10克中草药浓缩成1克的水提粉剂)对鳖源致病性嗜水气单胞菌株ZHYYZ-1(GenBank登录号:GU563992)进行体外抑制实验。采用倍比稀释法和平板法测定各药液对体外培养的嗜水气单胞菌的MIC及MBC值,为金银花1.56mg/ml、黄芩1.56mg/ml、蒲公英3.12mg/ml,并分别记做1个单方药物浓度单位。通过测定(1A,0.8A+0.2B,0.6A+0.4B,0.4A+0.6B,0.2A+0.8B,1B)(例如黄芩与金银花进行复配,则1A=1h=1.56mg/ml,1B=1j=1.56mg/ml)单方两两复配组合对实验菌的杀灭作用,
3、筛选出对实验菌具明显抑菌效果的蒲公英与黄芩组合(0.2p+0.8h、0.4p+0.6h、0.6p+0.4h,依次记为A、B、C)和黄芩与金银花组合(0.4h+0.6j、0.6h+0.4j、0.8h+0.2j,依次记为D、E、F),开展A、B、C、D、E、F间(例如A与D复配,则A=0.2p+0.8h,D=0.4h+0.6j,1p=3.12mg/ml,1h=1.56mg/ml,1j=1.56mg/ml)两两复配组合的杀菌效果评价实验。结果表明,(0.08p+0.60h+0.32j)与(0.16p+0.60h+0.24j)这两个复配组合杀菌率均达到99%以上,故将其作为治
4、疗淡水养殖动物嗜水气单胞菌病重症的中药配方,为进一步获取预防和治疗轻度的淡水养殖动物嗜水气单胞菌病的用量,再设置菌浓度为1×103cfu/ml、1×104cfu/ml、1×105cfu/ml三个组别,并相应稀释【(0.08p+0.60h+0.32j)、(0.16p+0.60h+0.24j)】复配组合药物浓度,采用最小杀菌浓度测定方法,确定其用量范围。实验表明,稀释倍数为达4倍时其杀菌率仍可达90%以上。[关键词]黄芩金银花蒲公英嗜水气单胞菌ZHYYZ-19ChineseHerbalMedicineonturtlePathogenicAeromonashydrophil
