4、ftendinopathy.TheeffectofWnt3aontheosteogenicdifferentiationoftendon-derivedstemcells(TDSCs)wasalsoexamined.Methods.Ossifiedfailedtendonhealingwasinducedbytheinjectionofcollagenaseintothepatellartendonofrats.Atvarioustimesthetendonswereharvestedforimmunohistoch
5、emicalstainingofWnt3a,b-catenin,Lrp5andTcf1.Patellartendonsampleswereobtainedfrom13patientswithpatellartendino-pathy(11unossifiedand2ossified)and10controls.ImmunohistochemicalstainingofWnt3a,b-catenin,Lrp5andTcf1wassimilarlyperformed.RatpatellarTDSCsweretreated
6、withWnt3a.TheosteogenicdifferentiationofTDSCswasexaminedbyALPactivity,alizarinredSstainingandmRNAexpressionofosteogenicmarkers.atZhejiangUniversityonNovember30,2013Results.TherewasincreasedexpressionofWnt3a,b-catenin,Lrp5andTcf1inthehealingfibroblast-likecells,
7、chondrocyte-likecellsandossifieddepositsintheanimalmodelandinsomeclinicalsamplesoftendinopathy.Wnt3aincreasedALPactivity,calciumnoduleformationandexpressionofosteogenicmarkersinTDSCs.Conclusion.ActivationoftheWntsignallingpathwayanditseffectonTDSCsmightcontribu
