3、),andtyrosine(Tyr)residuesisoneofthemajorpost-translationalmodificationsinthebacteria,involvedinregulatingamyriadofphysiologicalprocesses.Cyanobacteriaareoneofthelargestgroupsofbacteriaandaretheonlyprokaryotescapableofoxygenicphotosynthesis.Manycyanobacteriastrainscon
4、tainunusuallyhighnumbersofproteinkinasesandphosphataseswithspecificityonSer,Thr,andTyrresidues.However,onlyafewdozenphosphorylationsitesincyanobacteriaareknown,presentingamajorobstacleforfurtherunderstandingtheregulatoryrolesofreversiblephosphorylationinthisgroupofbac
5、teria.Inthisstudy,wecarriedoutaglobalandsite-specificphosphoproteomicanalysisonthemodelcyanobacteriumSynechococcussp.PCC7002.Intotal,280phosphopeptidesand410phosphorylationsitesfrom245Synechococcussp.PCC7002proteinswereidentifiedthroughthecombineduseofprotein/peptidepr
6、efractionation,TiO2enrichment,andLC−MS/MSanalysis.Theidentifiedphosphoproteinswerefunctionallycategorizedintoaninteractionmapandfoundtobeinvolvedinvariousbiologicalprocessessuchastwo-componentsignalingpathwayandphotosynthesis.Ourdataprovidethefirstglobalsurveyofphospho
7、rylationincyanobacteriabyusingaphosphoproteomicapproachandsuggestawide-rangingregulatoryscopeofthismodification.TheprovideddatasetmayhelprevealthephysiologicalfunctionsunderlyingSer/Thr/Tyrphosphorylationandfacilitatetheelucidationoftheentiresignalingnetworksincyanoba
