2、(第三军医大学:1医学检验系临床检验学教研室,重庆400038,2学员旅七队,重庆400038)摘要目的探讨以AEAPS及葡聚糖共修饰超顺磁纳米颗粒(superparamagneticnanoparticles,SPION)的制备方法,并检测其物理和磁学性质。方法共沉淀一步法制备葡聚糖包被的四氧化三铁纳米颗粒,然后在通氮、60℃、强力机械搅拌的条件下加入AEAPS,得到AEAPS与葡聚糖共修饰的SPION。采用透射电镜测量其大小及分布,用振动样品磁强计检测磁化率等参数,用普鲁士染色的方法判断AEAPS/葡聚糖-SPI
3、ON是否与抗体连接。结果所得样品核心为四氧化三铁晶体,核心粒径不超过20nm,包被葡聚糖后整体颗粒直径不超过25nm,质量饱和磁场强度为67.36488emu/gFe,AEAPS/葡聚糖-SPION与抗体结合,可对特异性抗原产生抗原-抗体结合,普鲁士染色结果为阳性。结论制备的样品具有粒径小,分散性好,超顺磁性等优点,能与单克隆抗体有效结合。【关键词】超顺磁性氧化铁;AEAPS;纳米粒子;抗体偶联PreparationandperformancetestforAEAPS/Dextran-modifiedSPIONDEN
