2、ossiblesignalingmechanismsbywhichrecombinanthumanplatelet2derivedgrowthfactor(rhPDGF)acceleratedhealingofcutaneouswoundindiabeticrats.MethodsFourfull2thicknessskinwoundswereincisedinthebackof26maleWistardiabeticrats.Thewoundedratsweredividedinto3groups(7or8ratseachgroup).Onegroupwithouttreat

3、mentwasusedasacontrol,andtheother2groupsweretreatedwith2rhPDGFatadoseof710Lgöcmwoundorvehicle(DMSOö019%NaCl,volövol1∶1)from1to14days.Thewoundhealingwasevaluatedbythemeasurementsofthewoundvolumeandarea.Immunofluorescentandimmunohistochemicalstainingwereusedtoexaminethephosphorylationofextrace

4、llularsignal2regulatedkinase1ö2(ERK1ö2)andtheexpressionofproliferativecellnuclearantigen(PCNA),respectively.ResultsGranulationtissueappearedinthebedofwoundafterinjury.ThenumberofbloodcapillarybudsandfibroblastswasgreaterintherhPDGF2treatedgroupthanthatintheother2groups.Alotofinflammatorycell

5、sinfiltrationandcollagendepositionwereobservedinthewound.Thewound2volumeintherhPDGF2treatedgroupwassmallerthanthatincontrolgroup(P<0.05).ThereepithelializationrateinrhPDGF2treatedgroupwashigherthanthatintheother2groupsat7daysafterinjury(P<0105).TheexpressionofPCNAinreparativecellswashigherin

6、rhPDGF2treatedgroupthanincontrolgrouporvehicle2treatedgroupat3,7daysafterinjury(P<0105).ThephosphorylationofERK1ö2wasstrongerinrhPDGF2treatedgroupthanthatincontrolgrouporvehiclegroupat7and14daysafterinjury(P<0105).ConclusionTheseresultssuggestthatrhPDGFaccelerateswoundhealingandimprovesheali
