3、vesasgroundworkforgermplasmimprovement.LaboratoryfocusedonscreeningmicrosatelliteprimersandanalyzinggeneticstructureamongHdiversicolorpopulationsinordertocapturesomeSSRprimersaswellastheoptimumamplificationconditionthatsuppliesfeasibleSSRmarkersandreferencefor
4、furthergeneticbreedingofH.diversicolor.1.PrimerScreening:themixedDNAtemplateof衄diversicolorWasemployedtoscreen83paksofSSRprimersandoptimizetheconditionofreaction.Asaresult,80pairsofutilizableSSRmarkershadcomeavailableforuse.Threepopulationswith30abaloneindivid
5、ualseachgatheredfromSanya,BeihaiandLinggaowereamplifiedundertheoptimalreactionconditionof80pairsofSSRmarkersjusttoyield66pairsofvalidmicrosatellite10ci.2.Analysisonpopulationgeneticstructure:111eaverage嫱ofthree豆diversicolorpopulationsofvariousgeographicalbackg
6、roundsWas0.1076withintermediategeneticdifferentiation.TheamountofallelesfromSanya,BeihaiandLinggaopopulationswere3.6,3.7and3.6respectively,accompaniedbyexpectedheterozygosity(HE)0.45614,0.48423and0.49343aswellasobservedheterozygosity(HO)0.39647,0.40000and0.404
