2、tionsofPGinplantresistanceanddiseasesresponseswerespeciallydiscussedinthispaper.ThemultiformityofPGfunctionswasindicated.Agrobacterium·-mediatedtransformationhadbeenusedtointroducetheAnti-PGgeneintoprocessingtomato(cv.98—75).Fourpositivetransformedtomatoplantswer

3、econfirmedbyPCRandSouthernhybridization.Fruitfirmnessandsolublesolidoftransgenictomatowerehigherthanthoseofcontrol;fruitwerefullandlustrous;solublepectinwaslowerthancontrolatthesamestage.ThePGactivityoftransgenictomatowas83%,88%,63%,46%ofcontroltomatorespectively

4、NorthernhybridizationshowedthatallPG,LOgandLeEXPlmRNAdecreasedintransgenictomatofruitcomparedwiththewildtomato.Theseresultssuggestthattheprocessofripeningandsofteningintransgenicanti-PGtomatoweredelayed,TheexpressionpatternsofPG.LeEXPlandLOXwerestudiedinwildtomat

5、o,antisenseACStomatoandNrmutanttomato.TheresultshowedthatPGplayedamajorroleinripeningandsofteningoftomatofruit,andcooperatedwiththeregulationofLeEXPlandLOX.TherelationshipbetweenathyleneandPolygalacturonase俨(甜Wasthoroughlyinvestigatedintwoaspectsoftheeffectofethy

6、leneonPGfunctionandtheregulationofPGonethylenesynthesis.Withwildtomato,antisenseACStomato,Nrmutanttomatoandwildtomatotreatedwithexogenousethyleneand1-MCPasmaterials,itwasshowedthattheexpressionofPGgeneandPGactivitywereinhibitedsignificantlyinantisenseACStomatoand

7、Nrmutanttomato.TheexpressionofPGwaspromotedwithexogenousethylene(100p.1/L)andobviouslyinhibitedwithl—MCP(1I_tl/L).TheseresultssuggestedthatexpressionofPGgenewasregulatedbyethylene.Ontheotherhand,withantisensePGtomatoandwildtomatotreatedwith100mg/LD-GAasmaterials,
