3、ificity,andtheemer-genceofdrug-resistantcellsubpopulationsoftenhampertheefficacyofdrugthera-pies.Despitethesignificantprogressachievedbychemotherapyinthetreatmentofdisseminatedmalignancies,theprognosisforsolidtumorsremainspoor.Anumberofspecificdifficultiesareassociatedwiththet
4、reatmentofsolidtumors,wheretheaccessofdrugstocancercellsisoftenlimitedbypoor,unequalvascularizationandareasofnecrosis.Thehistologicalheterogeneityofthecellpopulationwithinthetumorisan-othermajordrawback.Attemptstotargettherapiestotumorshavebeenaddressedbyusingprodrugsactivated
5、intumorsbyelevatedselectiveenzymesandaredescribedinChapter27.AnalternativestrategythatuseantibodiestotargettumorswithforeignenzymesthatsubsequentlyactivateprodrugsisdescribedinChapter26.Oneapproachaimedatenhancingtheselectivityofcancerchemotherapyforsolidtumorsreliesontheappli
6、cationofgenetherapytechnologies.Genetherapiesaretech-niquesformodifyingthecellulargenomefortherapeuticbenefit.Incancergenetherapy,bothmalignantandnonmalignantcellsmaybesuitabletargets.Thepossibil-ityofrenderingcancercellsmoresensitivetodrugsortoxinsbyintroducing“suicidegenes”h
