1、摘要目的本实验探讨藏红花素对骨髓造血干/祖细胞体外集落形成的影响。方法提取骨髓单个核细胞进行集落培养,分为空白对照组及藏红花素组,藏红花素浓度分别为0.25mg/ml、0.59/ml、lmg/ml、2mg/ml,培养14天后在倒置显微镜下进行总造血集落、红系造血集落计数。结果(1)藏红花素浓度O.25.1mg/ml时随藏红花素浓度增加总造血集落计数增加,总造血集落计数与藏红花素浓度成正相关,藏红花素浓度为lmg/ml时增加最显著;藏红花素浓度增加至2mg/ml时总集落计数减少;增加至4mg/ml时不形成集落。(2)藏红花素浓度在
2、0.25.1mg/ml时以粒一巨噬细胞集落(CFU—GM)、混合集落(CFU.GEMM)等非红系集落形成为主。(3)藏红花素浓度在1-2mg/ml时,随浓度增加总造血集落形成显著下降而红系集落形成增加。结论(1)在0.25.1mg/ml,藏红花素可以显著增加总集落及红系集落的形成,集落百分比以非红系为主;(2)在1.2mg/ml,藏红花素则降低总集落形成,促进红系集落形成;(3)适宜浓度的藏红花素可提高骨髓造血干/祖细胞集落形成,提示藏红花素能提高骨髓造血能力。硕士研究生丁雯雯儿科学指导教师孙立荣教授关键词:藏红花素;造血干/祖
