1、提要本文以丝素蛋白作为药物载体,采用W/0/W型复乳法,枝接抗凝血药物阿司匹林,研究抗凝血丝素缓释微球的制备;并对红花黄色素的溶媒法提取和初步纯化工艺进行了研究。研究内容包括:1、再生丝素溶液的制备;2、阿司匹林-丝素缓释微球的制备及质量评价;3、红花的提取纯化工艺及红花提取物微球的初步研究。探讨了乳化剂比例、丝素溶液的浓度、油水相比例以及阿司匹林与丝素的比例对丝素载药微球的制备及其性能的影响。采用紫外分光光度法测定丝素载药微球的载药率和包埋率,测定微球的释药性能,并且对载药微球的稳定性进行评价。以羟基红花黄色素A的含量为指标,采用单因素试验法考察了纤
2、维素酶浓度、提取时间、提取温度、pH等因素对红花黄色素提取量的影响;正交优化纤维素酶提取红花黄色素的最佳工艺;采用醇沉工艺对红花黄色素初步纯化,通过正交试验和单因素试验考察乙醇的质量百分数、搅拌速度、溶液的pH对醇沉后羟基红花黄色素A纯度及转移率的影响,经过初步纯化,羟基红花黄色素A的纯度可达50%左右。关键词丝素;阿司匹林;红花;纤维素酶。StudiesonPreparationofAspirinSilkFibroinMicrospheresandsilkfibroinmicrospheresofSafflowerSpeciaLity:Pharmac

6、Drugreleasepropertiesofmicrosphereswasevaluatedbyreleaseinvitro.WiththecontentofhydroxysaffloryellowAasindex,cellulaseconcentration,extractiontimeandtemperature,aswellaspHwerestudiedontheeffectofsaffloweryellowextraction.Theoptimizationofcellulasewasselectedbymeansoforthogonalte
