1、重庆医科大学硕士学位论文云芝糖肽对人PBMC中TLR信号通路调控机制的研究姓名:陈楚申请学位级别:硕士专业:免疫学指导教师:鲍依衡201205重庆医科大学硕士研究生学位论文。云芝糖肽对人PBMC中TLR信号通路调控机制的研究摘要目的:通过对云芝糖肽(PSP)作用人外周血单个核细胞(PBMC)后,TLR信号通路中关键节点基因的表达和关键蛋白的表达的研究,明确PSP对TLR信号通路的免疫调控作用,探索PSP的免疫调控和免疫抗肿瘤机制。方法:体外分离、培养人PBMC,分为对照组矛NPSP处理组,植物血凝素(PHA)诱导增
2、殖,采用实时荧光定量PCR(Q.PCR)技术对人PBMC中TLR信号通路中相关基因的表达差异情况进行定量分析,同时对TLR通路中关键节点采用免疫蛋白印迹法(Western—blot)检测其蛋白水平表达情况。结果:PSP处理盼实验纽与对照组相比较,PBMC的TLR信号通路中的TLR4、TLR5、TLR6、IRAK4、TRAF6、TAKl、IKKa、JNK、P38、ERK、IRF5和NF.KB基因的mRNA表达明显增高(P<0.05)。同时通过Western—blot检测TRAF6、IRF5、NF.1(B和TAKI蛋白
3、的表达均有明显增高(P<0.05)。结论:PSP的免疫调控作用可能主要通过TLR通路中NF—KB矛DIRF5等转录因子调控终端效应因子的释放来实现。与此同时,TLR通路中关键节点基因TRAF6的激活,可能通过TAKl进一步激活MAPK途径,重庆医科大学硕士研究生学位论文影响细胞的周期、生长、凋亡和细胞因子的分泌,为进一步探讨PSP的免疫调节作用机理提供了实验依据。关键词:云芝糖肽;TLR;免疫调节;Q—PCR4重庆医科大学硕士研究生学位论文REGULAToRYEFFECTSoFPOIⅣSACCHAROPEPTIDE
