1、青岛大学硕士学位论文海马--下丘脑弓状核Ghrelin神经通路构成及对胃运动的影响研究姓名:高海霞申请学位级别:硕士专业:病理学与病理生理学指导教师:徐珞20120529摘要目的:(1)探讨海马CM区与下丘脑弓状核ghrelin神经通路的构成;(2)研究ghrelin在海马CAl区一下丘脑弓状核通路对胃运动的调控。方法:(1)采用荧光金(FG)逆行追踪实验,观察下丘脑弓状核和海马间神经通路的构成,并结合荧光免疫组织化学方法观察ghrelin神经元在海马CAl区一下丘脑弓状核间的投射。(2)大鼠海马CAl区内微量注射ghrelin和ghrelin受体阻断剂D
2、-Lys3-GHRP-6,对清醒自由活动大鼠胃运动的影响。(3)电损毁下丘脑弓状核,观察海马CAl区微量注射ghrelin对清醒自由活动大鼠胃运动的影响。结果:(1)逆行追踪结合免疫荧光组化研究显示,海马与弓状核分别注射FG,7天后可见弓状核、海马有大量标记FG的神经元,结合ghrelin抗血清免疫组化实验,可见在Arc、海马FG标记的神经元细胞中有ghrelin免疫阳性物的表达。(2)在体胃运动研究显示,海马CAl区内微量注射ghrelin,可明显增强清醒大鼠胃运动(P0
