1、青岛大学硕士学位论文LBA对单眼视觉剥夺性弱视大鼠视网膜的保护作用姓名:刘相和申请学位级别:硕士专业:人体解剖与组织胚胎学指导教师:迟焕芳20090525LB^对单眼视觉剥夺性弱视大鼠视网膜的保护作用中文摘要目的建立单眼视觉剥夺性弱视大鼠动物模型,观察视觉剥夺性弱视大鼠视网膜的改变情况;观察枸杞子提取液(LBA)对视觉剥夺性弱视大鼠视网膜的保护作用,并探讨可能的作用机制,为弱视的临床治疗提供理论依据及可备选药物。方法新生Wistar大鼠21只,雌雄不限。随机分为对照组(5只)、实验组(8只)及治疗
2、组(8只)。1周后,应用褥式缝合法缝合实验组及治疗组大鼠单侧眼睑,建立单眼视觉剥夺性弱视动物模型。4周后,拆除实验组及治疗组手术缝线,并开始经口给予治疗组大鼠LBAlmg/kg灌胃治疗,每日一次。对照组及实验组自由进食。所有大鼠均在自然光线环境中饲养。6周后处死所有大鼠,摘取眼球。应用伊红一苏木素(rlE)染色观察视网膜的改变;应用TUNEL检测视网膜细胞凋亡的表达;应用免疫组织化学的方法,检测bcl.2蛋白(bcl.2)、BAX蛋白(BAx)及Caspase.3的表达。结果OnE染色可见对照组视
