1、原发性IgA肾病中微小RNA的差异表达研究眭维国,蓝慧娟,晏强,邹贵勉,李飞,郑艺花,黄河,陈洁晶,戴勇(中国人民解放军第181医院肾脏科,广西桂林541002)摘要:目的借助微小RNA(microRNA,miRNA)芯片研究IgA肾病(IgAnephropathy,IgAN)中miRNA差异表达。方法收集11例IgAN患者肾穿标本作为IgAN组,3例镜下病理检查正常的肾皮质作为对照组。使用Trizol提取每组总RNA,分离miRNA,采用miRNA芯片筛选出差异表达显著的miRNA,实时定量PCR技术验证芯片结果可靠性。结果IgAN中miRNA出现显著差异表达65个,其中
2、显著表达上调34个,显著表达下调31个。挑选hsa-miR-637、hsa-miR-492进行实时定量PCR相对定量检测,结果显示2种方法检测的miRNA表达量和表达趋势相符,芯片结果具有可靠性。结论IgAN是一种经典的多因素、多基因病,目前其机制尚未明确。本实验表明miRNA在IgAN中出现显著差异表达,深入研究将有助于IgAN机理研究及其诊断、治疗和预防。关键词:微小RNA表达;微小RNA芯片;IgA肾病中图法分类号:文献标识码:ThestudiesofMicroRNAsdifferentialexpressioninIgAnephropathyWeiguoSui,Hu
