1、天津医科大学硕士学位论文NF-κB在脂卵巢切除小鼠牙槽骨表达及其对成骨能力的影响姓名:张丰伟申请学位级别:硕士专业:口腔医学;口腔临床医学指导教师:贾智201105天津医科大学硕士学位论文中文摘要目的:为了探寻雌激素缺乏所致的骨质疏松症牙槽骨成骨能力与NF-r,B的活化之间是否存在内在的联系,从而为骨质疏松症患者牙周炎的治疗提供研究基础。本实验通过卵巢切除建立雌激素缺乏小鼠模型,运用组织学方法观察卵巢切除小鼠组织病理学改变,逆转录PCR检测小鼠牙槽骨中成骨相关标记物的表达情况,以及免疫组化法和免疫印迹法检测小鼠牙槽骨中核转录因子r
2、,B(NF.rd3)的活化情况和表达量的不同。方法:1.取20只3月龄雌性C3H的小鼠,随机分为两组:假手术组(A组)和卵巢切除组(B组)。切除双侧卵巢(或假手术),建立小鼠雌激素缺乏小鼠模型;2.10周后,断颈处死两组小鼠。取小鼠牙槽骨,经过固定、脱钙、包埋、切片和HE染色等步骤,观察小鼠组织学差异;3.处死后,立即取小鼠牙槽骨,剔除软组织后,’提取总RNA,经过逆转录、PCR和电泳等过程,检测OCN和Runx2等成骨相关分子mRNA的表达;4.采用免疫组化方法,观察两组小鼠牙槽骨中NF.心蛋白表达情况;5.应用免疫印迹法,经蛋
