1、血清蛋白质谱模型对胃腺癌诊断的应用性研究姓名：2014年6月25日血清蛋白质谱模型对胃腺癌诊断的应用性研究【摘要】［目的］探讨用蛋白质芯片技术筛选胃腺癌患者血清蛋白质表达谱，寻找血清中的标志性蛋白。［方法］采用蛋白质生物芯片表而增强激光解析电离飞行吋间质谱(SELDI)技术，运用SAX2(StrongAnionicExchanger)蛋白质芯片检测胃腺癌患者、胃炎患者和健旗者血清,建立诊断模型，然后进行争盲模型验证。［结果］发现5910Da、5084Da和8691Da的三个蛋白质荷比峰(M/Z)在胃腺癌和健康组比较屮具有显著性差异。5910Da、6440Da的两个蛋白

2、质荷比峰(M/Z)在胃腺癌和胃炎组中比较K宥显著性差异。［结论］建立了胃腺癌的血清蛋白指纹质谱，为以后的胃癌蛋白质组学研究奠定了一定的基础，建立了以5910Da、5084Da、8691Da和6440Da四个蛋白质峰为模型区分胃腺癌与非胃腺癌的血清蛋0表达质谱诊断模型，为胃腺癌的临床诊断提供了一条崭新的途径和方法。【关键词】表面增强激光解析电离-飞行时间质谱蛋0质芯片生物标记胃腺癌ApplicationofScrumProtcomicPatternsforDetectionofGastricAdenocarcinomaserumproteinprofilingofast

7、可达60%〜80%,但假阳性率较高，难以用于早期诊断。冈此，寻求早期诊断胃腺癌的特异方法是口前临床亟待解决的难题。我们利用表面加强激光解吸离子化飞行时间质谱（surface?鄄enhancedlaserdesorptionandionisation,SELDI）技术，对胃腺癌患者与正常人和胃炎患者血清中的蛋a质进行对比分析，筛选可用于临床胃腺癌早期诊断的特异性生物标记物。1材料与方法l.i血清样品所有病人血清样品均为2003年8月〜2004年6月台州市立医院手术后病理检查确诊为胃腺癌患者的术前抽检标本，对照样品来自经健康查体正常的健康志愿者及经病理检
