3、glia,theimmunecompetentcellsofthecentralnervoussystem(CNS),normallyexistinarestingstatecharacterizedbyaramifiedmorphology,andbecomeactivatedinresponsetobraininjury,infection,andavarietyofneuroinflammatorystimuli.Microglialactivationcaninducebeneficialordeleteriousph
4、enotypes,dependingonthecontextofactivation.Beneficialphenotypes(M2)aremediatedthroughanti-inflammatorycytokines,suchasIL-4andTGF-β.Incontrast,microgliacanbeacutelyactivatedbyLPSorIFN-γ,andreleasepro-inflammatorycytokinessuchasIL-6andTNF-a.Theregulationofmicrogliaish
5、ighlycomplex,dependingonmultifactorialsystems,includingtheneurochemicalmilieu.Studieshaveshownthatstimulationofreceptorsinresponsetoneurotransmitters,neuropeptides,andneuromodulatorsaffectmicroglialfunctions,includingcytokinereleaseandphagocytosisinresponsetoinjury.
6、Inthisstudy,wecomparedtheresponsivenessofmicrogliaindifferentactivationstagestoneurotransmitters.Activatedmicrogliaviastarvationtreatmentapparentlyshoweddifferentresponsestoserotonin(5-HT)ordopamine(DA).ManysignalingpathwaysincludingERK1/2areknowntobeactivatedby5-HT
7、,butERK1/2activationwasnotsignificantlyalteredupon5-HTorDAstimulation.However,wefoundasignificantincreaseoftheactivityofp38MAPkinaseinresting,butnotinactivatedmicroglia.TheincreaseofpaxillinphosphorylationatSer83byp38MAPkinaseupon5-HTorDAstimulationhasbeenalsoobserv
8、ed.Theactivationofphagocytosisbypaxillinandcofilinissuggestedbythefindingsthatphagocytosispositivelycorrelateswiththeactivationofbothpaxil
