ChemicalanalysisSophisticatedtoolsareavailableforstudyingthestructuralcompositionofbacteria(mostcommonlyfattyacid,carbohydrateorubiquinoneprofiling)ofbacteria.Characterizationofsecretedmetabolicproducts(e.g.volatilealcoholsandshortchainfattyacids)isalsohelpful. ApproachestorapiddiagnosiswithoutpriorcultureCertainhumanpathogenseithercannotbeisolatedinthelaboratorygrowextremelypoorly.Successfulisolationcanbeslowandinsomeinstancesimpossible.3.Directdetectionofbacteriawithoutcultureispossibleforsomeorganisms. AsimpleapproachtorapiddiagnosisAntigendetectionUsedinmanydoctor'sofficesforthegroupAstreptococcus.Thepatient'sthroatisswabbedandstreptococcalantigenextracteddirectlyfromtheswab(withoutpriorbacteriologicalculture).Thebacterialantigenisdetectedbyaggregation(agglutination)ofantibodycoatedlatexbeads. BacterialDNAsequencesamplifieddirectlyfromhumanbodyfluidsThepolymerasechainreaction(PCR)canbeusedtoamplifylargeamountsofspecificgenesorportionsofgenes.Greatsuccesshasbeenachievedinrapiddiagnosisoftuberculosis. DirectmicroscopicobservationCertainclinicalsamplessuchassputumcanbedirectlyassessedforthepresenceofbacteriacanbehelpful(e.g.detectionofM.tuberculosisinsputum). SerologicidentificationSerologicidentificationofanantibodyresponse(inpatient'sserum)totheinfectingagentcanonlybesuccessfulseveralweeksafteraninfectionhasoccurred.
