3、PRGFs)inOAsynovialcellbiology.Methods.Synovialcellswereisolatedfrom10osteoarthriticpatientsandculturedinserum-freemedia(basalconditions)andexposedtoeitheraplatelet-poorpreparationorPRGFfor72h.Cellsactivatedwithinterleukin-1(IL-1)for48hwerealsoexposedtoPRGF.Changesinseveralevent

4、srelevanttojointhomeostasisincluding(i)hyaluronicacid(HA)secretion,(ii)thebalancebetweenmetalloproteinase-1,-3and-13(MMP-1,MMP-3andMMP-13)andtissueinhibitor-1(TIMP-1)and(iii)thesecretionoftransforminggrowthfactor-1(TGF-1),vascularendothelialgrowthfactor(VEGF)andhepatocytegrowth

5、factor(HGF),wereallassessed.Results.PRGFsignificantlyenhancedHAsecretioncomparedwithplatelet-poorpreparations,P<0.05;atthesametimereleaseofTIMP-1,MMP-1,MMP-3andMMP-13werenotaffected.AnincreasedHGFproductionwasobserved(P<0.05)butVEGFandTGF-1levelsremainedunchanged.PRGFsignifican

6、tlyenhancedthesecretionofHAinducedbyIL-1activation,P<0.05,butitdidnotmodifytheDownloadedfromIL-1-inducedriseinMMP-1,MMP-3andVEGF.Incontrast,PRGF-inducedHGFproductionwasabolishedbythepresenceofIL-1duringPRGFtreatment,P<0.05.Conclusions.Intra-articularadministrationofPRGFmightbeb
