1、神经肽Y及其受体在实验性大鼠垂体腺瘤中的表达陈来照马景鑑基金项目:国家自然科学基金资助项目(30471772)作者单位:030001太原,山西医科大学第一医院神经外科(陈来照),天津医科大学总医院神经外科(马景鑑)通讯作者:陈来照(E-mail:chenlaizhao@yahoo.com.cn)【摘要】目的研究神经肽Y及其受体在实验大鼠垂体腺瘤中的表达,探索NPY在垂体腺瘤形成过程中的作用。方法雌性Wistar大鼠40只,通过腹腔注射雌激素的方法建立大鼠垂体腺瘤模型,在此基础上采用放射免疫法测定大鼠血浆NPY的浓度,应用免疫组化、免疫电镜和逆转录聚合酶链反应(R
2、T-PCR)的方法分别研究大鼠垂体中NPY及其受体的表达。结果(1)实验组大鼠垂体符合垂体腺瘤的诊断标准;(2)免疫组化和RT-PCR研究表明,在实验组大鼠垂体腺瘤中NPY及其Y1R和Y2R的基因表达水平明显低于对照组垂体。(3)免疫电镜显示NPY阳性颗粒主要位于腺垂体细胞的内分泌颗粒中。结论通过雌激素诱导Wistar大鼠,可以成功建立垂体腺瘤动物模型;实验组大鼠垂体腺瘤中NPY及其受体的表达下调可能与肿瘤的发生有关。【关键词】大鼠;垂体腺瘤;神经肽Y;受体。ExpressionofneuropeptideY(NPY)aswellasitsreceptorsin
